[EN] Panoramic optical mapping is the primary method for imaging electrophysiological activity from the entire outer surface of Langendorff-perfused hearts. The use of the de deckere-ten hoor preparation for study of nicotinic and potassium-evoked dopamine ?-hydroxylase release from the rabbit heart

Dopamine β-hydroxylase (DBH) and noradrenaline were determined both in the venous effluent (perfusate) and in the transmyocardial fluid (TMF) collected from the apex of the rabbit isolated heart which was prepared according to De Deckere and Ten Hoor (1977) and perfused with Tyrode's solution at 20 ml/min. 2. Perfusion for 2 min with the nicotinic drug, p-aminophenethyl-trimethylammonium (PAPETA), both in the absence of presence of atropine evoked noradrenaline overflow into the perfusate and TMF that was maximal in the 0–2 min sample and declined from maximum with a t1/2 of 0.6 min. The effect of dopamine on the overflow of endogenous noradrenaline from the perfused rabbit heart evoked by sympathetic nerve stimulation.

The effects of dopamine and two dopamine receptor antagonists (pimozide, flupenthixol) on the release of endogenous noradrenaline evoked by electrical stimulation of the postganglionic sympathetic nerves and their influence on cardiac performance were measured in isolated perfused rabbit hearts. 2. Dopamine 0.2μM decreased noradrenaline overflow and ventricular tension development in response to nerve stimulation. 3. The effect of methacholine on noradrenaline release from the rabbit heart perfused with indometacin

The experiments were undertaken in order to study the effect of inhibition of prostaglandin synthesis on the muscarinic inhibition of noradrenaline release evoked by sympathetic nerve stimulation. The right sympathetic nerves of the perfused rabbit heart were stimulated electrically. The noradrenaline output was enhanced after perfusion of the hearts with indometacin 3×10−5 M indicating blockade of the prostaglandin-mediated negative feedback control. Both in the presence and in the absence of indometacin methacholine 4×10−5 M decreased the noradrenaline output by a similar percentage. The experiments were carried out in order to investigate the functional significance of substitution of noradrenaline by amines known to exert actions weaker than those of the natural transmitter. On the isolated rabbit heart the potency relative to noradrenaline of (±)-dihydroxyephedrine to increase the rate was 1/4 and the potency to augment the contractile amplitude was 1/6. The corresponding values for (±)-dihydroxypseudoephedrine were 1/67 and 1/140, respectively.

Introduction and objectives. The analysis of frequency variability during ventricular fibrillation has yielded inconsistent results. We used an experimental model of ventricular fibrillation, with a short timescale, to analyze variations in frequency and their associated spatial distribution. Methods. A study is made of the characteristics of the atrial potentials recorded in the Koch triangle and its proximity, their variations on modifying the site of cardiac pacing, and their usefulness as markers of a distinct zone of the AV junction. In 12 isolated and perfused rabbit heart preparations an analysis was made of the endocardial atrial electrograms recorded with a multiple electrode positioned in the AV junction.
